Scope
"Introduction Ames assays of petroleum products have tended to give erratic results due mainly to the incompatibility of the test material with the aqueous bioassay system. This study was designed to determine whether by the application of minor modifications of the standard Ames assay protocol@ valid reproducible mutagenicity data could be obtained for petroleum products which have been assayed previously by other laboratories. The workscope was defined in three steps: 1. Conduct of a standard Ames assay on six petroleum materials according to Arthur D. Little@ Inc.'s protocols but making Individual 2 ml tubes of top agar components rather than taking 2 ml aliquots from 10 ml of a mixture of the common components. 2. Conduct of vortexing/sonication studies to develop an optimum mixing time and incorporation of these variables in the assay protocol to test the same six samples. 3. Assay of the same six samples utilizing a variation of the ""Pre-incubation"" assay. a. Pre-incubation of test materials (5 times final concentrations of bacteria@ enzymes@ test material) at 37??for 2 hours followed by a wash and plating in the normal test concentration (1/5 of incubation mixture)@ or b. Pre-incubation for 30 minutes with plating of the pre-incubation mixture containing test material. This report presents results of five petroleum materials tested as in Steps 1 - 3@ results of control assays designed to select optimum conditions for the conduct of Step 3 and results of the testing of the sample Identified as PS8-80D1 which was tested as a gas.
