Scope
Quality control testing of whole-genome amplified DNA products mainly includes two aspects: concentration testing and electrophoresis testing. The DNA product must have a concentration of not less than 30ng/μL and a total amount of more than 1μg after Qubit detection before sequencing analysis can continue; the electrophoresis band range of the DNA product must be more than 250bp and the upper limit is not more than 2000bp, and the main band is 750bp. After the samples pass the quality control test, they enter the library construction. The starting amount for library construction is 300ng. Currently, NEB double-stranded DNA fragmentation enzyme is used to fragment the main band into small fragments of DNA of 150-200bp. Subsequently, purification is performed, and the purified DNA is subjected to end repair, adapter ligation, PCR amplification, and purified library construction. After the library passes the BMG quantitative test, it will be sequenced on the computer.
